Molecular Cloning of Rat Liver and Testis Full-length cDNA Library and Isolation of Lactate Dehydrogenase A- and C- cDNA


Abstract
The glycolytic enzyme lactate denydrogenase(LDH) is a tetrameric protein composed of a specific combination of different subunits: A, B and C. These three isozynies have conservative aminoacid sequences and their genes are believed to be originated from an ancestral gene. The expression of these genes, however, shows tissue specificity and seems to be regulated independently at the transcriptional leveL And because LDH-A and-C subunits may thus be regulated at the transcriptional level, we have attempted to clone their cDNA with which isolate and characterize their genomic DNA sequences. We have constructed cDNA libraries with purified poly (A^(+)) mRNA isolated from rat liver and testes and screened to isolate cloned LDH A? and C-eDNA. The size of cloned LDH A- and C-cDNA were found to be 1.6kb and 1.1kb respectively and were partially characterized using several restriction eudonucleases.
